In Palermo airport terminal the bus ticket office is near the exit with two desks marked Segesta Lines for Trapani port or Prestia Comande for Palermo city. Buy your bus ticket here. (Sometimes one can buy the ticket on the bus but this is not guaranteed ... if you have time it is better to buy the ticket at the desk in the airport)
Outside the airport terminal turn right and walk to the end of the building (3 minutes) to the bus stop area.
Note that depending on the time of day, if you miss the Segesta airport bus from Palermo airport to Trapani, you can take the Prestia Comande city bus headed for Palermo and get off at Via Belgio as the bus enters the outskirts of the city; there you cross the road and walk 100 metres west along Via Belgio to the Segesta bus stop on the other side and catch the Segesta city bus as it leaves Palermo outskirts headed for Trapani.
